GMP-grade nanoparticle targeted to nucleolin downregulates tumor molecular signature, blocking growth and invasion, at low systemic exposure

Patients with breast or ovarian cancer have not benefited from improved efficacy pegylated liposomal doxorubicin relative to free drug, likely due the limited extent of enhanced permeability and retention (EPR) effect, further compromising drug bioavailability in tumor. Herein it is hypothesized that targeting nucleolin overexpressed tumor endothelial cells (readily accessible vascular compartment), besides cells, PEGASEMP (doxorubicin hydrochloride a lipid-based nanoparticle functionalized 31-aminoacid peptide nucleolin), lessens dependence on high systemic exposures EPR effect for successful targeting. This strategy has resulted intracellular doxorubicin, at low exposure, associated safe toxicological profile. Levels cell surface dictated antitumor activity against nucleolin-overexpressing solid tumors diverse histological origin, evidencing significant growth inhibition malignant mesothelioma over standard care. Those observations were paralleled by an impairment nucleolin-positive vasculature downregulation typically genes. Patient stratification based mRNA expression correlated prognosis enabled identification may potentially benefit PEGASEMP.
